There's an inherent contradiction there - you want creating a Manila site to be open to "anyone at anytime," except that you don't want it to be open to anyone (that is, including spammers) at any time.
So you don't want to block Web site creation - you only want to block /automated/ Web site creation, and now I finally read your subject line correctly.
Hasn't someone created an anti-comment-spam plug-in that displays letters and numbers in a skewed image that you have to type in to complete the process? That would seem to do the trick.
