Yes,
If you create an icon (.ico) file called favicon.ico and place it at /favicon.ico it will not generate a 404 error.

There are several shareware and freeware ico/icon editors that will allow you to create this file.

David Bayly's ManilaFixer plugin allows you to easily upload the favicon.ico file to your manila site.
